Аннотация:The terminal segment of the Bacillus FTU respiratory chain has been studied. The shapes of the difference spectra and the biphasity of the cyanide titration curve of respiration are suggestive of the presence of two terminal oxidases (aa3- and o-type) in the subcellular membrane vesicles of this Bacillus species. The aa3-type oxidase was shown to dominate at the beginning of the logarithmic phase, while the o-type oxidase was predominant at the stationary phase of bacterial growth. Both enzymes oxidized TMPD in the presence of ascorbate: the o-type oxidase with Km = 0.7 mM and Vmax = 2.2 micromol O2 min(-1) nmol(-1) haem o and the aa3-type oxidase with Km = 2 mM and Vmax = 11.2 micromol O2 min(-1) nmol(-1) haem a. KCN inhibited the oxidases: the half-maximal inhibition was observed at 2 microM and 20 microM for the aa3- and o-type oxidases, respectively.
